摘要:为了丰富烟叶细菌群落结构与多样性信息,更好地挖掘烟叶中有益微生物资源,以津巴布韦和我国四川普格、广东南雄及重庆彭水四地自然醇化相同时间的烟叶样本为材料,利用Illumina NovaSeq 6000测序平台对烟叶中细菌16S rDNA 进行高通量测序分析。结果表明,不同产地烟叶中细菌群落组成和多样性存在差异,津巴布韦和我国四川普格、广东南雄、重庆彭水四地烟叶样本分别获得827、1 381、458、246个OTUs,共有OTUs仅15个;烟叶细菌群落的丰富度和多样性在四地烟叶中都无显著差异(P>0.05),他们在四川普格烟叶中最高,其次为津巴布韦和我国广东南雄烟叶,重庆彭水烟叶中最低;变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)是烟叶中的优势菌门,假单胞菌属(Pseudomonas)、鞘单胞菌属(Sphingomonas)和芽孢杆菌属(Bacillus)是烟叶中的优势菌属。

Analysis of Bacterial Community Composition and Diversity of Mellowed Tobacco Leaves of Different Origins Based on High-throughput Sequencing

Abstract:In order to enrich the information on bacterial community structure and diversity of tobacco leaves and better explore the beneficial microbial resources in tobacco leaves, high-throughput sequencing analysis of bacterial 16S rDNA in tobacco leaves was carried out by using Illumina NovaSeq 6000 sequencing platform and tobacco samples from Zimbabwe, Puge Sichuan, Nanxiong Guangdong and Pengshui Chongqing of the same natural alcoholization time. The results showed that there were differences in the composition and diversity of bacterial communities in tobacco leaves from different production areas, and 827, 1 381, 458 and 246 OTUs were obtained from Zimbabwe, Puge Sichuan, Nanxiong Guangdong and Pengshui Chongqing, respectively, with a total of only 15 OTUs. The richness and diversity of tobacco bacterial communities did not differ significantly in the four tobacco leaves (P>0.05), and they were the highest among Puge Sichuan tobacco leaves, followed by Zimbabwe and Nanxiong Guangdong tobacco leaves, and Pengshui Chongqing tobacco leaves were the lowest. Proteobacteria, Firmicutes, and Actinobacteria are the dominant phylum in tobacco leaves, and Pseudomonas, Sphingomonas, and Bacillus are the dominant genus in tobacco leaves.
